The accident occurred around 4am on the Bucketts Way in Belbora, around 15 kilometres west of Krambach, when an ambulance and a vehicle collided.
The vehicle caught on fire as a result of the impact.
Police have reported that the ambulance was responding to a call and travelling east on the Bucketts Way before the head-on collision occurred with a vehicle travelling west.
According to police the vehicle travelling west had veered onto the incorrect side of the road prior to the impact.
The driver of the ambulance was airlifted to John Hunter Hospital in a serious but stable condition and the passenger was taken to Manning Hospital with non-life threatening injures.
NSW Ambulance has reported that both paramedics have since been released from hospital.
An off-duty Gloucester police officer was the first on the scene and assisted the driver of the vehicle.
The 38-year-old male driver had been thrown from the vehicle on impact.
He was taken to Manning Hospital in a critical condition before being transported to John Hunter Hospital.
The accident resulted in the Bucketts Way being closed to traffic for several hours.
The incident is being investigated by the police Crash Investigation Unit.
