Gloucester went into the match minus a couple of key players, but made the Ratz work hard for the points.
The Ratz support play improved in the second half and this saw them run in three tries, while their defence was strong throughout.
Second rower Emma Croker was outstanding for Manning as she constantly looked to put players into space with some clever passes.
The Ratz will play Old Bar Clams next Saturday at Taree Rugby Park while Gloucester will host Forster Dolphins.
In the other women's matches Wauchope thumped the struggling Forster-Tuncurry 94-14 while Wallamba cruised past Old Bar 55-5. Ellie Johnston scored five tries for Wauchope.
